My Number One tip I have, is to use the transparency tool to lighten whatever sketch you have scanned into illustrator. Before discovering this trick, I would trace over top of a full scan and I can't tell you how dumb I felt when I first discovered this trick. Oh well, you live you learn.
Other than that little tidbit, I use the pen tool and basically outlive the sketch and making adjustments as I go.
